ADF/thioredoxinによる発ガン遺伝子発現の制御

研究概要

HTLV-1感染細胞株培養上清より分離精製されたATL由来因子(ADF)は大腸菌の還元補酵素thioredoxin(TRX)のヒトhomologueであり、その活性中心に存在する二つのCys-SH残基を介した強い還元活性により種々の生物学手活性を要す。本年度はADF/TRXを含めた細胞内酸化還元調節因子と細胞内シグナル伝達機構との関連について解析を行った。ADFによる転写因子(NF-kB)の活性化については昨年度の本研究によりIL-2R/p55あるいはHI鹿のkb motifを介した転写がADF/TRXにより亢進することことを明かにしているが、AP-1に関してはT.Curranらの解析により、ADFがref-1蛋白を介してAP-1内のCysを還元することによりAP-1を活性化することが明かとなっている。そこで今年度は我々の系を用いてADF/TRX及びref-1蛋白のNF-kBへの影響を検討した。その結果ref-1蛋白がNF-kBの活性化にも作用していることが明かとなった。また細胞内酸化還元状態の細胞内蛋白チロシン燐酸化に及ぼす影響について検討を行い、酸化的ストレスが細胞内蛋白の顕著なチロシン燐酸化を誘導することを観察した。これらの結果は酸化還元状態と細胞内シグナ伝達機構との間にcross-talkが存在することを示唆している。
ADF/TRXはHTLV-1感染細胞のみならず、EBVやHPVによりトランスフォームした細胞や細組にも強い発現が認められることから、ADF/TRXのウイルス発癌への関与が強く示唆されている。本年度はウイルス発癌におけるADFの高発現の意義について解析を行うためにADF/TRXシステムの特異的阻害剤であるretinoic acid(RA)を用いた検討を行った。その結果RAによりADF高発現細胞株の増殖がADF低発現細胞細胞株に比べ選択手に抑制された。この結果はADF/TRXシステムの特異手阻害剤を用いたウイルス発癌の抑性あるいは治療への応用の可能性を示唆している。
