B細胞シグナル伝達機構の解析

研究概要	本研究計画でB細胞シグナル伝達を抗MB-1抗体を用いて解析を行なった。その結果MB-1分子は明らかにB系リンパ細胞に、positiveなシグナルを伝達い、Ca2^+濃度の上昇、チロシンリン酸化活性の上昇を誘導することが明らかになった。しかし、この反応は、成熟B細胞よりもむしろ、未熟なB前駆細胞に顕著に認められた。また、MB-1分子はlgMレセプターを発現する以前のpro-B細胞やpre-b細胞で発現しているのみならずヒトの細胞ではlg遺伝子、T細胞レセプター遺伝子の再構成が全く見られない。リンパ系前駆細胞でも発現しているこどか明らかになった。これらの結果は、この分子は成熟B細胞ではlgMレセプターの発現とシグナル伝達に関与しているが一方、未熟なB前駆細胞でも何らかの重要な機能を有していることを示唆しいてる。これらのB細胞の分化ステージでのMB-1分子の機能を明らかにすることによって、B細胞の初期分化、活性化への糸口となると考え、新たにMB-1分子に結合する機能分子の解析を行なった。MB-1分子とともに共沈される分子量52kDaのタンパクを同定し、これに対するモノクローナル抗体を作成した。この分子はチロシンキナーゼを含む活性化分子群とともに沈降されることが明らかになった。現在この分子のB細胞活性化における役割を解析するとともに、この分子をコードすると思われるcDNA遺伝子クローニング、塩基配例の解析を行なっている。
