正常骨髄造血幹細胞への試験管内遺伝子導入による免疫細胞調節遺伝子の研究

研究概要

リンパ系細胞の増殖分化に関わると考えられる分子の数は現在増大の一途をたどっている。しかし、抗体やリガンドを用いてその機能を検討できる細胞表面上のプロセスと異なり、細胞内の分子の機能について正常細胞を用いて検討することは現在もなお困難である。この問題の一つの解決方法として、我々は長期骨髄細胞培養法とレトロビールスベクターを用いる遺伝子導入法とを組み合わせることで、様々な遺伝子を正常B細胞分子化の培養系に導入することを試みた。本年は、培養やビールス感染方法等の改良を行ない、遺伝子導入の効率を上げるための諸条件を検討した。ベクターについてはこれまでpZip-neo,pGD-neo,pM5-neoの3種類について検討した結果、pM5-neoが長期骨髄培養系への感染では優れていた。しかし、現在もなお新たなベクターの開発が続けられているので今後もこの点については随時検討していく必要がある。次に、パッケージャー細胞の調整の仕方については、これまでよく行なわれている最初からstabel transformantを得る方法はベクター内に挿入した遺伝子の安定性に問たがあることがわかり、transient transfectionと感染を組み合わせた方法が最も問題が少ないことがわかった。一方、骨髄細胞培養側については、ストローマ細胞株とSCFを組み合わせて使うことで、造血幹細胞の増殖効率が格段に促進することを見いだし、この条件でビールスの感染、および遺伝子導入後の選択を行なっている。このような条件で、c-mosやLacZ等の遺伝子をpM5-neoに挿入してビールスを調整し、長期骨髄細胞培養系への遺伝子導入を行なったところ、培養中で感染、遺伝子導入細胞の選択、そしてこの細胞からのB系細胞の誘導を全て行なえることが明らかになり、初期の目的はほぼ達成することができた。
