研究概要

最近の私共や他の研究室の解析により、ras p21が遺伝子発現系を介して細胞の増殖や分化を制御していることが明らかになりつつある。本年度は、ras p21の標的蛋白質の同定および活性化機構について解析を行い、以下の結果を得た。最近、ras p21が細胞レベルでMAPキナーゼを活性化することが報告されているが、本年度、私共はツメガエル卵の細胞質画分にKi-ras p21に依存してMAPキナーゼを活性化する因子が存在することを明らかにし、これをREKSと命名して部分精製した。REKは分子量150kDaの蛋白質で、細胞質に存在していた。ras p21はそのC末端に脂質による翻訳後修飾を受けることが知られているが、Ki-ras p21がREKSを活性化するのにこの翻訳後修飾が必須であった。私共はすでに、smg GDS(GDS解離促進蛋白質)がKi-ras p21のGDP/GTP交換反応を促進して活性化することを見出して報告しているが、最近ras p21に作用する新たなGDSとしてGRFが他の研究室によって見出されている。本年度は、これら二種類のGDSによるras p21の活性化機構を比較解析した。smg GDSとGRFは同一条件下でほぼ同程度の初速度でKi-ras p21のGDP/GTP交換反応を促進した。私共のこれまでの解析から、smg GDSは翻訳後修飾を受けているras p21にのみ作用することが明らかとなっているが、GRFも翻訳後修飾を受けているras p21のほうにより高い活性を示した。GRFはsmg GDSが有する今一つの活性であるKi-ras p21を細胞膜から遊離させる活性も有していた。本年度の私共の研究結果から、ras p21の標的蛋白質と活性制御機構について着実な成果をあげることができ、本年度の研究計画はほぼ達成することができたと考えている。
