虚血性ニューロン壊死に関わる興奮性アミノ酸とカルシウムの動態

研究概要	虚血侵襲に脆弱な海馬CA1ニューロンが、虚血に誘発されて遅発性ニューロン死をおこす初期のメカニズムを分析した。 1)スナネズミを用い、in vivoマイクロダイアリシスを行うと両側総頚動脈結紮により海馬CA1領域に急速大量のグルタミン酸放出がおこったが、この現象は領象非選択的であった。このグルタミン酸に由来は、シナプス前部、ニューロン胞体、アストログリア細胞の三部位にわたり、シナプス前部においても代謝型グルタミン酸プールからの放出であることも明らかとなった。 2)スナネズミ海馬スライスを用い、カルシウムマイクロフルオロメトリーによって虚血負荷時のニューロン内カルシウム濃度[Ca^<2+>]iの動態を検索した。虚血シミュレーションを負荷すると一定潜時の後、CA1領域に限局した[Ca^<2+>]i上昇がおこった。この上昇の起源は1/3が細胞外からの流入、残り2/3はリヤノジン型とIP_3型が同程度寄与する細胞内貯蔵からの放出であることがわかった。予め一過性虚血を負荷した動物から採取したスライス実験から、虚血負荷が何らかのメカニズムによりグルタミン酸受容体や細胞内カルシウムチャネルの持続的活性化をおこしていることも明らかとなった。 3)興奮性アミノ酸神経毒性を惹起する特異的リガンド、AMPAやキスカル酸の海馬膜標品結合実験では、従来のオートラジオグラフィーによる報告と異なり、亜部位間の受容体密度差に顕著なものは見られなかった。 4)ラット胎仔の海馬よりアストログリアの初代培養を行い、虚血シミュレーションによるグルタミン酸、アスパラギン酸のグリア放出機構の存在を明らかにした。
