研究概要

吉田:ミトコンドリア外膜と内膜のコンタクトサイトに存在し、マトリックスや膜間腔蛋白質の輸送装置の一員である29kDa蛋白質の抗体は、ポリンの輸送定着をも阻害した。従って、本輸送装置は外膜蛋白質の輸送にとっても必須であることが知られた。森:前駆体蛋白質のミトコンドリア移行を促進するサイトソル因子を大腸菌に発現させ、精製し、抗体作成と機能解析を進めている。更に、hps70が輸送に必要であることをin vitroの系で証明した。伊藤:シトクロムb5の部分融合蛋白質の細胞内分布の解析から、b5の小胞体残留シグナルは、膜挿入部に存在することが分かった。ミトコンドリアプロセシングペプチダーゼの基質認識機構を合成ペプチドを用いて解析し、-2位のアルギニン残基が必須であることを示した。長谷:非光合成型フェレドキシンの光エネルギー依存の葉緑体移行過程において、前駆体分子の大部分は膜透過しているが延長ペプチドの切断は受けていない中間状態の存在、を見い出した。この状態は、暗所でのATP依存の過程では検出されない新規なものである。橋本:カタラーゼのペルオキシソームへの局在化機構を、変異体タンパク質を無カタラーゼ血症患者細胞内で発現させることにより検討した。局在化シグナルは他のマトリックス酵素と異なりペプチド内部に存在していた。また、ヘムの結合および、4量体形成はペルオキシソーム内で起こることを示唆する結果を得た。米田:セミインタクト細胞を用いたin vitro系において、精製核蛋白質であるヌクレオプラスミンが尿素処理してはじめて効率良く核内に移行することが明らかとなり、その移行効率にhsc70が関与していることを証明した。また、このin vitro系を利用して、hsc70以外の核蛋白質輸送に必要な因子の精製を進め、その候補分子を複数見つけている。
