研究概要

吉田:ラットミトコンドリア外膜と内膜のコンタクトサイトに存在し、マトリックス、外膜、膜間腔そして恐らくは内膜の蛋白質の輸送装置の一員である29kDa蛋白質のcDNAをクローニングした。その塩基配列の解析から本蛋白質はミトコンドリア蛋白質特有のプレ配列を有していることが判明した。森:動物におけるタンパク質のミトコンドリア移行におけるサイトソルのHsp70の役割を解析した。Hsp70ファミリーのうちHsc70が関与しており、前駆体タンパク質の膜透過の段階には必ずしも必要ではなく、合成途上の前駆体に結合して膜透過可能な状態に保つことが明らかとなった。伊藤:シトクロムb5の小胞体局在化にC端部での電荷数が、定着に膜挿入部の後半が関与していること、ミトコンドリアプロセシングペプチダーゼの基質認識に必要な共通構造として、切断部位から近位及び遠位に塩基性アミノ酸、、両者の間にグリシンやプロリンが存在することが必要であること、が解った。長谷:非光合成型のプラスチド蛋白質の葉緑体への輸送過程はストロマの酸化還元状態により調節されており、光照射下の還元状態では前駆体のままで蓄積することを明らかにした。また、この輸送経路は光合成型蛋白質とは異なることを、延長ペプチド領域の合成ペプチドを用いた輸送競合実験で示した。橋本:ペルオキシソーム膜タンパク質の局在化機構を明らかにするために、ペルオキシソームの内在性膜タンパク質の2つのABC輸送体ALDPとPMP70に関して、試験管内での膜へのインポート系を確立した。この系を用い、ALDPの種々の欠失変異体の膜へのインサーションを検討したところ、N-末端側の疎水性の領域が必要であることが明らかとなった。米田:核蛋白質輸送の最初のステップにおいて、核蛋白質を中心にして大きな複合体が細胞質で形成されて核膜にまで運ばれることを明らかにした。その複合体を構成する4つの因子のうち、必須の働きをする2つの因子について、蛋白質の完全精製に成功し、その部分アミノ酸配列から遺伝子クローニングした結果、1つは未知の分子であることがわかった。遠藤:アカパンカビMOM22のホモログとして、酵母ミトコンドリアでタンパク質の外膜透過に関与するMAS17を同定した。また、エンドウにおいてSecAホモログ遺伝子を同定し、その発現産物が葉緑体内部でタンパク質のチラコイドへの輸送に関与していることを証明した。
