研究概要

小胞体経路の各オルガネラの形成機構とオルガネラ間の蛋白質の選別輸送機構を研究代表者と研究分担者で分担して研究を進め、下記の研究成果を得た。成果の大半は既に論文として学会誌などに発表している。
1.小胞体:小胞体膜蛋白質P-450について、小胞体に組み込まれたシグナルアンカー配列のアミノ末端が内腔側に存在することを証明し、シグナルアンカー配列に続くプロリンに富む配列が小胞体に組み込まれたP-450分子の高次構造形成に重要であることも確かめた(大村)。小胞体膜の蛋白質膜透過に関与する膜成分を透過中のペプチドとの化学架橋により検討し、9kDaの膜蛋白質を確認した(大村)。ミクロゾーム型アルデヒド脱水素酵素のC末端の疎水性アミノ酸配列が、この酵素の小胞体膜への挿入シグナルとしてだけでなく小胞体への留置シグナルとしても機能することを確かめた(田代)。P-450のシグナルアンカー配列の膜貫通領域が小胞体への留置シグナルとして機能することを確かめた(大村)。
2.小胞体-ゴルジ装置間輸送:この輸送過程に関与するSarlpによるGTPの加水分解は小胞体からの輸送小胞の形成以降に必要であることを無細胞輸送系を用いて確認した。
3.ゴルジ装置:プロ型前駆体蛋白質のプロセシングを行なう酵素Furinのゴルジ装置局在化シグナルを検討し、膜貫通領域よりC末端側の細胞質に露出したペプチド中にあることを見いだした(池原)。
4.リソソーム:ラット肝リソソームの膜蛋白質である酸性ホスファターゼのリソソーム局在化シグナルは細胞質に露出している短かなペプチド中のGYモチーフと考えられているが、酵母細胞で発現させた場合にはGYモチーフを改変しても膜貫通ドメインさえあれば液胞膜に運ばれることが確かめられた。動物細胞と酵母では局在化シグナルが異なるようである(姫野)。カテプシン群の内在性阻害蛋白質であるシスタチンalphaの細胞内転送にCキナーゼによるリン酸化が関与することを確かめた(勝沼)。
5.ペルオキシソーム:誘導剤投与で増殖した肝細胞内のペルオキシソームの膜と基質にユビキチンが高い濃度で存在することを免疫電顕法で確認し、余分なペルオキシソームの分解にはユビキチン系が関与する可能性がある(横田)。
6.トランスサイトーシス:培養上皮細胞にLDL受容体を導入しLDL受容体の細胞内輸送の実験系を確立した(高野)。
