枯草菌における蛋白質分泌装置構成成分の構造と機能の解析

研究概要	枯草菌はα-アミラーゼやプロテアーゼなどの有用酵素を大量に分泌生産する非常に強いタンパク質分泌能力を持っている。本菌におけるタンパク質分泌機構を解析するため、分泌装置構成成分の構造と機能の解析をおこなった。主要構成成分と考えられているSecAタンパク質の遺伝子は枯草菌細胞分裂の温度感受性変異(div変異)を相補できる遺伝子として発見した。841アミノから成り、大腸菌SecAより60アミノ酸小さく、50%の相同性が認められた。またN末端付近1/4部分では70%以上もの相同性が認められた。枯草菌SecAに対するウサギ抗体は大腸菌SecAと弱いながらも交差した。これらの結果は2種類のSecAタンパク質が類似していることを示した。しかし枯草菌SecAの機能を大腸菌タンパク質分泌変異株を用いて調ベたところ、in vivoおよびin vitroの両系で作用しなかった。ところが驚くベきことに、N末端部分約300アミノ酸からなる欠失ペプチドを利用した場合には分泌タンパク質のプロセシングを回復させるだけなく、高温における細胞の増殖も回復させた。HPLCによるゲルロ過法で解析したところ、欠失ペプチドは細胞膜に結合して存在し、大腸菌MM52株の変異SecAの機能を回復していることを明らかにした。一方枯草菌には高等生物に存在することが確認されているシグナルペプチド認識粒子が存在しているか否かを調ベるために、枯草菌small cytoplasmic RNA,およびこのRNAに結合すると考えられているFfhタンパク質についてもそれらの遺伝子を単離し、構造および機能について解析した。
