研究概要	本年度は最終年度なので、これ迄の研究をとりまとめる方向で研究を進めたところ、2つの大きな成果を得た。ひとつは柴田を中心にしたアクチン調節蛋白グループの高橋との共同研究が動脈硬化の遺伝子治療への道が開かれたことである。高橋の発見になるカルポニンは分化した平滑筋にのみ発現し、動脈硬化層の内膜平滑筋には発現していないことがわかっていた。カルポニンcDNAを良好な発現プロテータにつけて、ウサギの実験動脈硬化作製のバルーン処理にひきつづき、リポゾールとカップルさせ高圧下でうちこんだところ、カルポニンcDNA注入なしでは動脈硬化が出現していたのに、カルポニンcDNA注入では硬化はなく、カルポニンが多く発現していた。これは動脈硬化抑制への遺伝子治療の第一歩であり注目される。一方野々村らはゲルゾリンファミリー74kDaが、成体頸動脈では発現していないのに、培養初代細胞では発現してくることを見出し、一方蛍光抗体法でみていくと、球状の異状な細胞にはじまり、ここから双極性のいわゆる平滑筋的な細胞が産出され、増殖し、いわゆるHill&Valloyを形成する。この時Hillの頂上に球状細胞も増殖しつつ存在していた。74kDaはこの球状細胞にのみ存在していた。一方頸動脈、大動脈の組織をみてみると74kDaは生れたてから内膜にのみ僅か存在しているが、内皮障碍、動脈硬化、内膜肥厚のある細胞では74kDa存在細胞が、内膜肥厚部を中心に中膜にもふえていた。この結果は、血管に幹細胞的なものゝ存在を示し、こゝから未分化型の動脈硬化で出現する平滑筋を産生すると考えられ、これは血管学一般にとって重大な発見である。他にミオシン膜蛋白でもいくつかの発展があった。
