研究概要	血管機能における神経並びに体液性因子の影響を血管内皮平滑筋連関に焦点をあてつつその分子機構を研究し病態生理的意義を明らかにすることを目的とする本班の第2年度の活動は順調に進展している。まず第1に特記すべき成果は血管内皮由来弛緩因子(EDRF)としてのC-typeナトリウム利尿ペプチド(CNP)の発見である(井村、中尾)。細胞質グアニル酸シクラーゼを活性化する一酸化窒素(NO)とは対照的にCNPはBタイプ膜型グアニル酸シクラーゼを活性化してcyclic GMPをsecond messengerとするナトリウム利尿ペプチドファミリーのひとつであり、初めて発見されたペプチド性EDRFとして今後の発展が期待される。またこのCNPは急性単球性白血病由来細胞株のTHP-1細胞をTPA処理した際にCNPが産生されることが証明され(南野)、この発見はマクロファージ系細胞がCNPを産生することを示唆している。CNPは、今後血管内皮平滑筋連関の重要因子になるものと期待される。エンドセリンに関する研究も進展し、ヒトのET_A受容体とET_B受容体遺伝子の全構造の決定と染色体の同定(ET_A受容体:第4染色体、ET_B受容体:第13染色体)に成功した(井村、中尾)。またウシのETb受容体遺伝子構造と発現調節メカニズムの解析(広瀬)、個体発生におけるエンドセリン遺伝子発現の検討(山田)、エンドセリンを産生するマウス血管内皮細胞F-2腫瘍株の確立(今村)が行なわれた。また、血管内皮平滑筋の収縮蛋白質のCa^<2+>感受性にある種のG蛋白質の関与が推定されたが、rho A p21がCa^<2+>感受性を調節していることを明らかにし(高井)、血管平滑筋の収縮弛緩機構における低分子量G蛋白質の作用機構の解明の進展が期待できる。 NO(EDRF)の臨床的意義に関する研究も進展し、腎での意義(猿田)、高血圧における意義(平田)、心不全や冠循環における意義(竹下、友池)が検討された。更に細胞障害性活性化マクロファージの放出する血管弛緩因子の存在も報告され注目された(河合)。リポ蛋白と血管内皮平滑筋連関についての研究も独創的な展開をみせており、内皮細胞におけるスカベンジャー受容体の発現(北)、変性LDLによる内皮依存性弛緩反応の低下(泰江)、トリグリセリドリパーゼの解析(山本)が行なわれた。更にNa^+/H^+逆輸送担体のトランスジェニックマウスの開発(永井)、血管内皮平滑筋連関におけるアクチビンの意義(小島)、血小板由来内皮細胞増殖因子(豊島)、脳血管内皮細胞の研究(加藤)も行なわれた。
