血管内皮細胞における機械受容チャネルの研究

研究概要	血管内皮細胞は、伸展刺激に応じて血管作動性物質の分泌や形態変化などの様々な生理応答を示すことが知られている。しかしその分子機構は良く分かっていない。本研究の目的は、既にパッチクランプ法により内皮細胞での存在が知られている伸展活性化チャネル(SAチャネル)が、こうした機械応答の受容体として作動しているか否かを明らかにすることである。昨年報告したように、我々はシリコン膜上に培養した内皮細胞を用いて、伸展刺激に伴う細胞内Ca動員を測定することに成功した。また種々の薬理実験に基づいて、このCa動員が,細胞膜上に分布するSAチャネルの活性化によるといういくつかの証拠を提出した。一方Morrisら(1991)は、細胞全体に伸展刺激を与えても、単一チャネル記録(パッチクランプ)から予想されるような伸展活性化電流が記録できないことから、細胞応答におけるSAチャネルの関与に疑問を投げかけている。しかし彼らの伸展刺激は定量的ではなく、その刺激がSAチャネルを活性化させるに充分であったか否かは分からない。またSAチャネルを活性化するのに必要な伸展刺激量(膜張力)は技術的困難のために今まで測定が困難であった。今回我々は、超高倍率ビデオ顕微鏡のもとでパッチクランプを行うことにより、電極内の微小膜(パッチ膜)に発生する膜張力を評価し、SAチャネルの活性化に必要な張力を推定した。またシリコン膜法で細胞を伸展したときに発生する膜張力を評価することにより細胞内Ca動員に必要な張力を推定し、パッチクランプの結果と比較した。その結果、両者は良い一致を示し、細胞への伸展刺激で生じるCa動がはSAチャネルの活性化に基づくという仮説がより強固なものとなった。
