研究概要

平成4年度には以下のような進展があった。
1.培養細胞において、液胞型ATP分解酵素の阻害剤バフィロマイシンの効果が調ベられた。バフィロマイシンはプロラクチンの細胞内輸送をトランスゴルジ綱様体ないし、分泌顆粒の成熟過程で阻害した。またバフィロマイシンは細胞の増殖を抑制し、S期の細胞を増加させることが明らかとなった。
2.液胞型ATP分解酵素のサブユニットBは腎臓型と普遍型の2種存在しており、両種ともMDBK細胞で発現していることが明らかとなった。
3.酵母液胞型ATP分解酵素の活性発現に必須な遺伝子VMA12の構造が決定された。本酵素のプロティン・スプライシングは、(1)Cys738における切断(2)Cys284における切断(3)新しく生じたC端とN端の再結合という3段階を経ることが明らかとなった。
4.古細菌には液胞型ATP分解酵素しか存在していないものと思われたが、一部のメタン菌にはF型ATP分解酵素が存在していることがわかった。
5.好塩性古細菌ハロバクテリアの液胞型ATP分解酵素の触媒頭部は硝酸イオンによって阻害されるが、本来の活性であるATP合成活性は同イオンによっては阻害されないことが判明した。
6.連鎖球菌のNa^+輸送性、液胞型ATP分解酵素のクローニングが進行した。
