研究概要

まず,真核細胞の液胞型ATPaseについて述べる.網膜色素上皮細胞における視細胞外節の貧食は,貧食胞の形成,リソソームとの融合,融合したファゴリソソームにおける分解の3つのステップより成るが,液胞型ATPaseの阻害剤であるバフィロマイシンはその最後のステップだけを阻害することが明らかになった.また液胞型ATPaseが抗ガン剤タウノマイシンの細胞外排出過程に関係していることも明らかになった.酵母の液胞型ATPaseについては,そのαサブユニットが自触媒的にプロテインスプライシングを受けて成熟することが明らかになり,そのメカニズムが提案された.また,液胞型ATPaseの分子集合および活性発現に必須の2種種の遺伝子が新たに同定された.原核生物の液胞型ATPaseについては連鎖球菌Na^+輸送性ATPaseのオペロン全長の塩基配列が決定された.11の遺伝子から成り,3つが疎水性,8つが親水性蛋白質であり,真核生物の液胞型ATPaseのサブユニットとよく対応することが明らかとなった.好塩菌,好熱菌の液胞型ATPaseの全遺伝子の決定も順調に進行中である.好熱菌の液胞型ATPaseについては,蛋白質としても精製標品が得られるようになり,8種類のサブユニットから成り,輸送するイオンはNa^+ではなくH^+であることが判明した.好塩菌の液胞型ATPaseについては,種々の細菌種について検討した結果,それぞれ活性に必要とされる塩の種類などに相当の変化があり,バラエティに富んだものであることが判明した.
