研究概要

生物においてエネルギー通貨としての役割を果たしているATPはミトコンドリア、葉緑体、および細菌に於て合成されている。すなわち電子伝達鎖の形成するH^+の電気化学的ポテンシャル差を駆動力としてATP合成酵素がADPとリン酸からATPを合成している。本研究ではATP合成酵素のF_1部分の高次構造、活性中心部分の構造とH^+輸送との共役機構、調節蛋白の構造と作用機構等を明らかにするべく研究を進めている。
本年度に得られた成果は以下のとおりである。
(1)大腸菌のATP合成酵素の活性中心サブユニットβに系統的に変異を導入し、Lys-155,Thr-156,Glu-181,Arg-182,が触媒残基であることを証明し、Glu-181がH_2O分子の活性化、Lys-155がADPのリン酸基に配位し、Thr-156がMg^<2+>結合部位となるモデルを提出した。(2)変異および戻り変異の解析からγサブユニットのN末端側とC末端側が相互作用しながらH^+のゲートとしての機能を持つことを明らかにした。(3)大腸菌ATP合成酵素(F_0F_1)の中性子小角散乱実験を行ない、構造に関する情報を得た。(4)本酵素のα_3β_3サブユニット複合体の結晶化実験を行ない3,7Aの反射を記録できた。(5)好熱菌のATP合成酵素のサブユニット間相互作用、特に4次構造変化とプロトンの吸収と放出に関し新しいモデルを提出できた。(6)プロトン輸送機構を酸化還元共役ポンプを中心に解析し関与するアミノ酸残基を明らかにした。(7)ヒト・ミトコンドリアの関連する遺伝子の発現制御機構を明らかにした。(8)ミトコンドリアの本酵素の逆反応を阻害しているインヒビター蛋白がαとβサブユニットの境界面、特にαのGly-244とAsn-434の間およびβのTyr-359の近傍に結合することを証明した。
このように本酵素の高次構造と反応機構の解明という究極の目的に向かって研究を進めている。
