研究概要

bcl複合体の基質還元反応でbは急速に還元、再酸化される。bは単なる酸化剤ではないと示唆された。結晶標品はb558と2種類の2Fe-2S中心を持つことが判明し、酵素が2量体で機能することを示唆している。(松原)ウシ心筋bc_l複合体の3次元結晶のX線回折と、2次元の電子線解析の18A分解能電子密度をもとに、3次元結晶中での本複合体分子の方位と並進量を決定し、低分解能の3次元結晶構造が得られた。(福山)近赤外用ラマン分光計を製作し、チトクロム酸化酵素のCu_A部位の分子構造の共鳴ラマンスペクトル測定に世界で初めて成功した。酸化酵素とcとの複合体形成やa_3-Cu_B部位の酸化還元状態の変化、リガンドの結合でCu_Aの配位構造は変化しないこと、Cu_A部位がほかの酸化還元部位から独立していることを示した。(小倉)回虫の発生段階で好気的代謝を行なう幼虫の複合体IIを酵素化学的に調べた。FRD活性はなく、SDHのみの哺乳類型に似ていた。クロマト溶出位置も成虫とは異なっていた(ミトコンドリア複合体IIアイソザイムの存在の初例)。大腸菌複合体IIの部位特異的変異解析からヘムbの配位に関与する2つのヒスチジンを同定した。(北)グラフィックスを中心とした蛋白質結晶のX線による構造解析系を構築し、ヒドロゲナーゼや硫酸還元菌のエネルギー代謝系に関与する蛋白質のX線回折測定、構造解析を行なった。ヒドロゲナーゼ結晶の解析で6A分解能の電子密度を得、鉄硫黄クラスターとニッケル原子の位置を同定できた。(安岡)ウシ心筋チトクロム酸化酵素の結晶化の再現性を妨害する要因は用いたコール酸中の微量不純物によることが明らかとなり、不純物除去法を検討している。分解能向上のため、界面活性剤の種類を検討し、3.5A分解能のX線回折を示す結晶を得たが結晶化の再現性が低いのでその向上に努力している。(吉川)NAD(P)Hのレーザー励起ラジカルによるチトクロムc還元機構を明らかにし、還元型cから酸化酵素へ供給された電子が、ヘムaと銅に迅速に分配される過程を実測し、電子伝達過程を定量的に解析した。この励起がプロトンをもパルス的に生成するのでプロトン輸送過程の解析への応用を図っている。人工リポソームに再編成したbcl複合体を用い、膜電位により駆動されるプロトンの流れが複合体内で電子逆流をもたらすことを証明した。(折井)
