二次性能動輸送系の遂次結合型エネルギー共役機構の熱力学

研究概要	本研究は、ほぼ計画通りに進行し、次の段階への足がかりができたと考えている。交付申請書の計画に従って述べる。 1.生理・生化学的研究(電気生理学的測定及び放射性同位元素を使用した輸送反応の測定から、遂次結合モデルの各種パラメーターを見積もる):研究費を設備備品費に割けなかったため、輸送反応の電気生理学的測定を行えなかったが、一方、従来から行っている輸送反応・基質結合反応の測定結果をまとめ、機械論的モデルにおける輸送反応の各種パラメータの見積りを行い、輸送反応を遂次結合モデルでうまく説明できることを示すことができた。この結果、遂次結合モデルの一般性を確立できたと考える。 2.生化学的研究:これまで精製してきた輸送タンパク質の結晶化のための安定性を検討した結果、大腸菌輸送タンパク質はこれまでの各種界面活性剤や条件では不安定であることが判明した。つまり、好熱菌等のより安定な輸送タンパク質の検索が必要であると考えられた。 3.分子生物学的研究:Na^+依存性輸送タンパン質のアミノ酸配列のホモロジー検索を行った結果、かなり多数のNa^+依存性輸送タンパク質にNa^+結合部位と考えられる共通の配列が見いだされた。しかし、そのうちの一つのアミノ酸への部位特異的変異導入により、輸送活性が消失する場合(R^<376>->K)もあるが、そうでない場合も合り、Na^+結合部位であるというよりそのアミノ酸が構造的に重要な役割を果たしていることが示唆された。今後さらに、これまで得られた各種変異の戻り変異等の検索をして、構造と機能の関係に付いての情報を拡充していくつもりである。
