孤束核ニューロンのシナプス伝達における酸化窒素の役割に関する研究:特に逆行性精報伝達物質としての可能性について

研究概要	近年中枢神経においてL-アルギニンからnitricoxide(NO)が産生されることが証明されているが、NOが交感神経活動詞節に関与しているか否か明らかでない。本研究では、ラット脳幹部スライス標本の孤束核神経細胞活動記録法を用いて、孤束核におけるNO産生機序とその作用を検討した。〔結果〕1)孤束核神経細胞の42%で、L-アルギニン投与により用量依存性にスパイク頻度が増加した。孤束核神経細胞の47%ではスパイク頻度が不変、11%では減少した。2)L-アルギニンに反応した神経細胞において、D-アルギニン投与ではスパイク頻度は増加しなかった。3)L-アルギニンによるスパイク増加は、L-アルギニンのanalogueであるNMMAにより用量依存性に阻害された。4)L-アルギニンによるスパイク増加は、guanylatecyclaseの阻害剤であるmethyleneblueにより阻害された。5)L-アルギニンによるスパイク増加は、細胞外でNOと結合するヘモグロビンにより阻害された。6)L-アルギニン投与に反応した神経細胞は、NOを産生するSNP投与によりスパイク頻度が増加し、L-アルギニンに無反応の神経細胞はSNP投与によりスパイク頻度が増加しなかった。7)L-アルギニンに反応した神経細胞はグルタミン酸にも同様に反応した。〔結論〕上記の成績は、1)孤束核神経細胞において、L-アルギニンからNOが産出されること、2)産出されたNOは、産生細胞自身に作用するのでなく、周辺の神経細胞のguanylatecyclaseを活性化してCGMPを増加させ、神経細胞活動を亢進されること、3)孤束核神経細胞の約1/2がNOに反応すること、を示すものである。我々はinvivoの研究において、孤束核にNMMAを微量投与すると血圧、脈拍、腎交感神経活動が亢進することを確認している。これらの成績は、孤束核による交感神経活動調節にNOが重要な役割を果していることを示唆する。
