研究概要

本年度は、9人の計画班員に4人の公募班員を加えて、それぞれが順調に成果を上げた.
スプライシンググループでは、RBD/RRM型のRNA結合性蛋白質をコードするゼブラフィッシュ遺伝子を同定した。また、ヒトRNAヘリカーゼ様因子HRH1に、高等真核生物のスプライシング因子の特徴の一つであるRSドメインが存在することを明らかにした。一方、グループIイントロンにおけるスプライシングのメカニズムについても生化学的な解析な進展した。tRNAグループにおいては、牛ミトコンドリアtRNA^<Ser>_<OGA>について、通常のtRNAに存在すると考えられるDアーム付近の2つのMg^<2+>結合部位が存在しないことがわかった。また、牛ミトコンドリアtRNA^<Ser>_<OGA>のアンチコドンアームとコドントリプレットとの相互作用に関する研究についても順調に進展している。一方、感度的にはRI法に匹敵するtRNAアミノ酸受容能の新しい非RI測定方法を開発した。第3のグループであるNMRグループでは、昨年度までに開発した安定同位体標識導入システムを活用し、グループIイントロンについて、基質グアノシンと認識部位との近接においてステム(P9.0)が重要であることを検証し、また、16SrRNAのデコーディング領域と抗生物質との相互作用の位置を特定した。さらに、グアニン結合アプタマ-の高次構造が、グアニンの結合にともなって大きく変化することを明らかにした。Sx1タンパク質についても、RBDIがRBDIIと同様に長いループ構造をもつこと、および、このループ部分のアミノ酸組成がこれまでに知られているRBDと異なり芳香族アミノ酸に富んでいることを明らかにし、さらに、その特定の芳香族アミノ酸が、RNA結合の特異性に重要であることを明らかにした。
