研究概要

研究の目的:光合成の効率に影響を与える環境要因としては、温度、塩、無機イオン欠乏などが挙げられるが、これまでの研究により、上記の環境因子への適応に関する多数の遺伝子が単離された。本年度は、これらの遺伝子を他の植物・藻類・シアノバクテリアに導入し、さらに作成された形質転換植物の表現型を解析することを目的とした。
主要研究成果:(1)低温適応・耐性:不飽和化酵素の遺伝子を順次破壊した形質転換シアノバクテリアを用いて、膜脂質の不飽和化が光化学系II複合体の光失活に伴うD1タンパク質再成を促進することを明らかにした。(2)塩耐性:塩耐性に関与するベタインを合成するコリンオキシダーゼの遺伝子(土壌細菌由来)をシアノバクテリアとシロイヌナズナに導入し、これらの生物を塩耐性ならびに低温耐性にすることができた。(3)乾燥と低温:シロイヌナズナのホスホイノシトールに特異的なホスファターゼCの遺伝子の発現は、乾燥と低温のいずれによっても誘導されることを明らかにした。(4)CO_2濃縮機構:CO_2濃縮能を欠損したシアノバクテリアを作成し、その相補を利用してCO_2濃縮酵素の遺伝子を単離した。
