形質転換植物を用いた光合成組織における窒素酸化物会謝応答の分子機構の解析

研究概要	本研究は、NO_2を"好んで"代謝し、必要な窒素源を大気中のNO_2に依存する"好NO_2植物"の育成を究極の目的とし、硝酸代謝関連酵素遺伝子をシロイヌナズナに導入したトランスジェニック植物を作成、本植物における導入した遺伝子の発現を分子レベルで解析し、以て植物葉におけるNO_2代謝応答の分子機構を解明することを直接の目的とする。本問季は以下の成果を挙げることができた。 1.道路沿いに生育する野生草本植物52種、栽培草本植物60種、栽培木本植物37種、合計149種についてNO_2同化能力を調査した結果、シロイヌナズナは6番目に高いNO_2同化能力を有することがわかった。また、シロイヌナズナを4±1ppmのNO_2ガスに8時間暴露すると、総還元態窒素の6%以上がNO_2由来の窒素で置き換わることが見いだされた。このことは、シロイヌナズナはNO_2を窒素源として利用する効率が高く、本研究の目的にふさよして実験植物であることも示するものである。 2.ホウレンソウNiR cDNAをCaMVプロモーターおよびNOSターミネーターに結合した発現ベクターを遺伝子銃法によりシロイヌナズナに導入し、除草剤耐性遺伝子をマーカーとしてトランスジェニック植物を作成した。これまでに、導入したホウレンソウNiR遺伝子がシロイヌナズナの染色体DNA上に組み込まれていることの確認(サザンハイブリ分析による)されたトランスジェニック植物を2系統(10個体)獲得した。 3.ホウレンソウNiR cDNAをプローブとしてシロイヌナズナゲノミックNiR遺伝子をクローニングについて研究し、シロイヌナズナゲノミックNiR遺伝子の全塩基配列を初めて決定した。この配列から予想されるアミノ酸配列はホウレンソウNiRのアミノ酸配列と約80%の相同性を有していた。
