研究概要

成長期のプロテアーゼ:液胞タンパク質を成熟させるプロセシング酵素のcDNAを初めて単離し構造解析を行った(西村)。LHCP IIのクロロフィルとアポタンパク質の量比調節に関わるプロテアーゼの性質を明らかにした(田中)。インゲンマメの発芽時特異的に発現するSHプロテアーゼ遺伝子の発現調節領域と相互作用を示す発芽時特異的な核タンパク質を見いだした(南川)。
成熟期のプロテアーゼ:光化学系II反応中心D1タンパク質のC末端プロセシング酵素を精製して構造解析を行った(佐藤)。酸素発生系の18-kDaタンパク質に特異的に作用するプロリルエンドプロテイナーゼを精製し性質を調べた(桑原)。植物の26Sプロテアソームを初めて精製し性質を調べた(中川)。易分解性FNRのN末端はブロックされていることを示した(新)。植物におけるカルパインの存在を検索した(鈴木)。D1タンパク質の光分解に対する界面活性剤の効果を調べた(池内)。ストロマのエンドペプチダーゼのクローニングを試みた(清田)。
老化期のプロテアーゼ:乾燥ストレスで誘導されるATP要求性プロテアーゼの調節サブユニットのcDNAをクローニングし構造解析した(篠崎)。コムギのRubiscoのin vivo分解産物を検出した(前)。
今年度の研究により、いくつかの植物細胞内プロテアーゼについて精製/クローニングが完了し、それらの特異的機能発現のメカニズムを明らかにする道が開けた。来年度は、これまでの研究により重点的に研究を進めるべきであると思われるプロテアーゼの発現調節および基質認識(基質タンパク質の分解されるべきフォームとされるべきでないフォームの識別)機構の解明に重点を置いた研究を進める。
