植物細胞内プロテアーゼの生化学的・分子生物学的手法による総合研究

研究概要	成長期のプロテアーゼ:液胞プロセシング酵素(西村)および発芽種子子葉のシステインプロテアーゼ(南川)について、不活性型プレプロ前駆体から活性型成熟体へのプロセシングを細胞内輸送の過程を明らかにした。光捕集クロロフイルタンパク質複合体形成時に、ラジカル発生の原因となる遊離のクロロフイルを防ぐための、アポタンパク質の過剰生産-余剰分解の「保険機構」を明らかにした(田中)。成熟期のプロテアーゼ:光化学系IIの反応中心D1タンパク質について、C末端プロセシング酵素の認識部位を解析し同酵素をクローニング(佐藤)、その光分解をin vivoで観察する系をらん藻で構築(池内)し、分解が光化学反応時に必然的に生じるヒドロキシルラジカルによることを明らかにした(徳富)。酸素発生系の23-kDaタンパク質について、局所的かつ可逆的なコンフォメーション変化とチラコイド膜結合プロテイナーゼに対する感受性の関係を明らかにした(桑原)。ストロマのエンドペプチダーゼをクローニングした(清田)。N末端のみが異なる2種類のFNRの生理的機能と昜分解特性の関係を調べた(新)。植物で初めて26Sプロテアソームを精製し立体構造を明らかにした(中川)。植物にはカルパインは検出されないが、プロテアソームやD-アミノ酸分解酵素は検出されることを示した(鈴木)。老化期のプロテアーゼ:道管形成時の細胞死に関わるプロテアーゼをクローニングした(福田)。乾燥(老化類似)ストレスに時に発現するシステインプロテアーゼ遺伝子の発現調節機構を調べた(篠崎)。植物の自然老化時におけるRubiscoの分解を解析した(前)。
