研究概要

今年度はまず平成4年度に得られた細胞内および細胞外の各種入力信号に応答するイオンチャンネルの分類と生理的役割の検討を以下のごとく行った。自律神経節細胞のMチャンネル(赤須、小林)、中枢神経細胞のイオノトロピックおよびメタボトロピック受容体刺激の電気的膜特性とそれに関連する細胞内2次メッセンジャー系の同定ならびに各受容器間の相互作用(赤池)、電位依存性Naチャンネル解放物質の構造特性(瀬山)、血管内皮細胞の内因性Kチャンネル活性化物質の生理的役割(鈴木)、平滑筋(富田)及び心筋(野間)細胞のイオン電流に対する物理的修飾機構、体温調節中枢の温度感受性ニューロン(清原)、小腸上皮細胞のメカノトランスダクションチャンネル(岡田)、イカ巨大神経軸索のClチャンネル(山岸)の電気生理学的分類と細胞内制御機構の同定、ミトコンドリア膜電位勾配の制御機構(北里)、細胞内Caシグナルのモニタリングとその放出・流入制御機構(井上,九場、反町)、神経分化の細胞内制御(高橋)、ギャップ結合の形成過程(外崎)、レセプターを介する軸索輸送の制御(竹中)。研究計画ではその上でさらに2次メッセンジャーを介したチャンネル・チャンネル間共同現象の発現メカニズムを解明することにしていたが、結果は必ずしも十分ではなかった。その原因は電気生理学的測定法の難しさにあると言って良い。たとえばイオンチャンネルの開閉を視覚的に正確に記録するためにはパッチクランプ法が不可欠であるが、それでは細胞内の2次メッセンジャー系は破壊されてしまう。ニスタチンはその有効な手助けとなるが、イオンチャネル開閉の解析はノイズ解析を用いざるを得なくなる。本研究の研究分担者はそれぞれの分野で日本を代表する専門家であり、今後ともさらに密に連絡を取り合い、問題点の克服と技術の普及に努める必要がある。
