免疫アジュバントを用いるヒトウイルスワクチンの免疫力増強

研究概要

近年、ウイルスワクチンの開発はリコンビナントワクチン、コンポーネントワクチンや合成ペプチドワクチンを中心に行なわれており、飛躍的な展開を見せている。しかしこれら合成ワクチンやリコンビナントワクチンは抗原性が弱く、有効な免疫アジュバントの開発が待たれる。本研究班ではムラミルジペプチド誘導体であるMDP-Lys(L18)とB30-MDPがサイトカイン誘発剤や、ワクチンの免疫アジュバントとして有効であることが見出された。特にB30-MDPはインフルエンザのコンポーネントワクチン、B型肝炎のリコンビナントワクチンに対して血中抗体産生、細胞性免疫の誘導に強いアジュバント活性を示した。MDP-Lys(L18)はアラムを含まないB型肝炎のリコンビナントワクチンに対して両者を単に混合するだけで強い抗体産生増強を示した。B30-MDPは不活性化ハンタウイルスワクチンおよびリコンビナント核タンパクに対して血中抗体産生および細胞性免疫の誘導に免疫アジュバント活性を示した。本班において昨年に引きつづき、本年もワクチンの免疫力増強に有効な新しい免疫アジュバントの開発が進められており、合成リピドA誘導体、合成DNA、細菌菌体成分のテイコ酸や糖脂質、人工多糖などの免疫アジュバント活性についても明らかにされた。ウイルスワクチンの実験系としてハンタウイルスの表面抗原及び核内抗原の組換えタンパクの作製に成功し、これらリコンビナントタンパクに対するB30-MDPの有効性を検討している。また、ヒト免疫不全ウイルス(HIV-1)、ヒトヘルペスウイルス6(HHV-6)の感染モデルや抗原分析も進められており、これらウイルス感染モデルに対する合成免疫アジュバントの応用も検討中である。
