吸入曝露における許容曝露限界値の決定方法に関する総合的研究

研究概要	吸入曝露における許容曝露限界値を決定する際に考慮すべき重要な問題点のうち、動物実験による経口毒性データから人の限界値を推定する際の基本的条件として、経口と吸入による吸収化学物質の代謝及び体内動態の差を明確にしておくことが重要であることをまづメタキシレンとクロロホルムをモデル物質として明らかにした。金属元素の毒性が元素の酸化度及び化学形によっていかに異なるかを水銀蒸気と無機水銀化合物をモデルとし、共存するセレン濃度との関連において明示し金属元素の許容曝露限界値は異なる酸化数の化学形ごとに個別に決定する必要があることを明らかにした。金属のうち、感作性を有する元素に対する許容曝露限界値の決定は特に困難な課題であるが、コバルトを含む超硬合金関連作業者について、特異的IgE値とライフスタイルの関係を解析し、この両者間に有意の関連を認めたことにより、ライフスタイルを含めた大きな個体差の存在を念頭に置いて限界値を設定する必要があることを示した。金属への曝露量を定量するには吸入空気中濃度と共に、血液、尿、などを用いる生物学的モニタリングが有用であるが、鉛をモデルとして影響指標によるモニタリングがきわめて有用であること示した。また指標の精度を高めるための精度管理の重要性を指摘した。有機溶剤による神経系への毒性を調べるための新しい方法を検討し、グリア細胞マーカーの有用性を明らかにした。有機溶媒は一般に混合曝露が多いが、トルエンとキシレンへの単独曝露と混合曝露を多数の作業者について比較検討し、混合曝露の際の許容濃度設定のための基礎データを提供し得た。更に感作性物質一般の許容濃度の設定について論じ、定量的な数値を示すことが困難な場合には、単に感作性物質表を提示することが有効であるとして、新たに感作性物質表を作成した。
