脳細胞機能・代謝・分子構築連関からみた軽度〜重度脳虚血とその治療的手段

研究概要

本年度は虚血脳の情報伝達系に関する基礎的研究として各種脳内レセプタ分子構築とその機能について検索を進めるとともに、虚血による中枢神経細胞障害を抑制する要因についてさらに検索を進めた。
脳虚血による神経細胞障害に密接に関連するとされるグルタメートレセプタについては、NMDAレセプタのサブタイプの構成によってプロテインキナーゼC(PKC)によるリン酸化反応に差異があり、この、PKCによるリン酸化を受けやすいサブタイプは大脳皮質、海馬等に分布していることがわかった。また、κ-オピオイドレセプタについてもクローニングが成功し、一次構造の決定がなされ、生理学的性質と構造の関連、また虚血病態との関連について、分子構築からの検索を進めることが可能となった。さらに、レセプタを介する細胞内情報伝達系に関連する蛋白について、とりわけ、シナプス前からの神経伝達物質放出機構に関与するカルシウム結合蛋白を同定した。これらカルシウム結合蛋白の作用と虚血による細胞内Ca^<2+>上昇との関連もさらに検討する必要があると考えられた。
一方、脳虚血による神経細胞障害に対する治療的手段として、NOの産生の抑制やアラキドン酸代謝系阻害の有効性について検討がなされ、NO産生阻害薬が脳虚血における梗塞相の領域を減少させること、アラキドン酸代謝系の阻害薬が低酸素+無グルコースによる海馬切片神経細胞の膜電位の不可逆性脱分極を抑制することがわかった。また、前年度から検討している脳損傷による低酸素+無グルコースによる細胞内Ca^<2+>上昇に対する抑制効果は、損傷1-3日後に強く現われ、損傷直後(2時間)や7日後以降では有意な効果が認められなかった。これらの薬物および内因性抗虚血性作用の機序を明かにすべく、作用物質の本態とその生理学的性質についてさらに検索を進める予定である。
