研究概要	本年度に得られた主な結果を以下に示す。 1)海馬切片用いた虚血性灌流モデル(低酸素+無グルコース)において、ニューロン膜の急峻な脱分極から1〜1.5分後に膜の不可逆性変化が生じることが示唆された。また、その不可逆性変化にはNMDA受容体からのCa^<2+>流入と細胞内サイトからのCa^<2+>放出の関与が示唆された。2)軽度のアシドーシスは低酸素+無グルコースによる急峻な脱分極を遅延すること、また、このときの細胞内Ca^<2+>上昇を抑制することを観察した。3)長期増強の発生する領域に限局したグリア細胞の細胞内Ca^<2+>濃度上昇が観測され、神経可塑性や虚血時におけるグリア細胞活動の重要性が示唆された。4)海馬苔状線維-CA3ニューロンの長期増強のメカニズムについてカルバコールの作用をホールセルクランプによって解析した結果、海馬苔状線維終末での長期増強はシナプス前性に誘導されることが示唆された。5)神経終末部がシナプス後膜に付着した単離ニューロンにおけるIPSCを指標として、化学伝達物質遊離に関与する電位依存性Ca^<2+>チャネルの性質を検索したところ、N型高閾値Ca^<2+>チャネルは殆ど関与せず、P型及びL型高閾値Ca^<2+>チャネルの寄与が大きいことがわかった。6)レセプタを介する細胞内情報伝達系について、シナプス前からの神経伝達物質放出機構に関与するカルシウム結合蛋白を同定した。これらカルシウム結合蛋白の作用と虚血による細胞内Ca^<2+>上昇との関連もさらに検討する必要がある。8)脳虚血の際に未知の遺伝子群の検索のため、極めて高効率に特定の遺伝子群を単離できる新しいcDNAサブトラクション法"Directional tag PCR subtraction法"を開発した。9)脳虚血による神経細胞障害に対する治療的手段に関連して、NO産生阻害薬が脳虚血における梗塞相の領域を減少させること、アラキドン酸代謝系の阻害薬が低酸素+無グルコースによる海馬切片神経細胞の膜電位の不可逆性脱分極を抑制することがわかった。
