研究概要

1.自然界からのC-P化合物の検索,分離と構造の決定に関する研究:アメフラシ神経組織の燐糖脂質のうち、約5〜10%は易水解性燐を含む糖脂質である。本研究に於て、非環元末端糖としてピルビン酸結合ガラクトースを持ったものの中に2種の新しい糖脂質を検出し構造を決定した。両糖脂質共、直鎖ガラクトースの6位のOHにフオスフオエタノールアミンを結合し、そのうちの1種は更に側鎖のガラクトースに2-AEPを結合したキメラ分子であった(佐武)。環形動物(ヒトツモンミミズ)の中性スフインゴ糖脂質中に、フオスファチヂールコリンを結合した2種の燐糖脂質を分離し構造を決めた。脂溶性部分も特異な構造を持つ(杉田)。アメフラシ卵の中に新しい2-AEP結合糖脂質を同定した。その構造からアメフラシ糖脂質群の生合成経路が示唆された(林)。2.C-P化合物の組織分布に関する研究:甲殻類(オキアミ)から分離したC-P糖脂質(MAE Pn Glc Cer)に対し抗血清を作成、抗体精製後免疫学的性質を明らかにし、オキアミの神経組織を特異的に染めることを明らかにした(杉田)。3.C-P化合物の生物活性に関する研究:アメフラシC-P糖資質がA-キナーゼの触媒サブユニットと結合し、A-キナーゼを活性化することを明らかにした。その他の蛋白質燐酸化酵素に対しては働かない。(佐武)。4.C-P化合物の生合成機構:抗生物質フオスフオマイシンのC-P結合生合成に重要なホスフオエノールピルビン酸ホスフオムターゼ及び次の反応であるPn Py脱炭酸酵素の遺伝子を、これ等酵素の欠損株を利用してクローニングすることに成功した(瀬戸)。5.C-P化合物の化学合成に関する研究:L-乳酸から(IS)-O-ベンジロキシプロオンアルピデヒドを合成し、後者を基質として立体選択的C-P結合を持つ1-ヒドロキシ生成物を合成した(山下)。
