研究概要

1.自然界からのC-P化合物の検索、分類と構造の決定に関する研究:アメフラシ(Aplysia kurodai)及びタツナミガイの卵からC-P糖脂質を分離し、構造を決定した。成体の糖脂質と異なり非還元末端にα3-0-メチルガラクトースを持ちN-アセチルガラクトサミンを欠き、3ケ(佐武)或は1ケ(林)の2-AEPを結合していた。アマクサアメフラシ(Aplysia juliana)の卵からグルコース1ケ、ガラクトース2ケと1ケの2-AEPを持つ新しい糖脂質を同定した(林)。2.C-P化合物の組織分布に関する研究:各々セラミド・アミノエチルホスホン酸(イケチョウガイ)、ホスホノグルコセレブロシド(南極オキアミ)を抗原として兎抗血清を作成し、抗体認識部位の同定、抗原の季節的変動、及び組織分布を明らかにした。抗ホスホノグルコセレブロシド抗体はテナガエビの神経組織を特異的に染めた(杉田)。3.C-P化合物の生物活性に関する研究:アメフラシ(A.kurodai)のホスホノ糖脂質はcAMP-依存性蛋白質燐酸化酵素を活性化する事が明らかになったが、糖鎖の違いによる活性化強度に大きな差は見られなかった(佐武)。4.C-P化合物の生合成機構:ビアラホスのC-P-C結合はホスフィン酸のP-メチル化反応で生じる。P-メチル化酵素遺伝子のクローニングにより本酵素蛋白が550ケのアミノ酸からなり、プロトポルフィリンを基質とする酵素とのホモロジーが認められた。従ってメチル供与体がメチルコバラミンである可能性が高い(瀬戸)。5.C-P化合物の化学合成:糖のヘミアセタール環内の酸素原子をリン原子で置き換えたリン糖化合物の合成を不飽和複素環化合物の2-ホスホレン類及び3-ホスホレン類に種々の官能基を置換して行い種々化合物の合成に成功した。更にこれらの新しい化合物に生理作用を見出し、医学的応用の可能性を示した(山下)。
