研究概要

1.全体としての成果ーー2回の班会議と1回の公開シンポジウムを開催し、そこでの議論から次の諸成果を得た。
(1)現状の把握ーー遺伝子診断が臨床医療の場で実用化されつつある状況に関して、広く情報を収集、交換した。
(2)相互の理解ーー法・倫理・社会学系と基礎・臨床・検査医学系の研究者間で知識と現状認識および研究的取り組みにギャップがあることを認め、相互ティーナインの必要性を確認した。
(3)問題点の整理ーー現実化しつつある遺伝子診断の臨床応用において上記ギャップの他に、検査技術の改良、データベースの充実、情報ネットワークや管理体制の整備など、多くの問題点があることを明らかにした。
(4)重点領域への申請ーー上記の議論を通じて、この問題に関する重点領域研究の必要性を総意として確認した。この総意に基づき、班員の提案を集約する形で申請書を作成し、平成6年度の重点領域に応募した。
2.個々の成果ーー全体としての成果の他に、班員はそれぞれの分野で以下(例示)の成果をあげた。
(1)遺伝子診断のための基礎検討ーー上田はアミロイド前駆体のmRNA発現がアルツハイマー病で異常なことを見出した。また、河合はIL8について、寺田は癌抑制遺伝子について、DNA診断のためのデータを集積した。
(2)DNA検査法の開発ーー大久保はPCRを用いた抗酸菌検査法を、榊は自動シーケンサーを用いたOrphan Peak Analysisを、また成澤はPCRを用いた中鎖アシルCoA脱水素酵素欠損症の簡易診断法を開発した。
(3)ソフトウェアの開発ーー清水はヒト遺伝子マップをデータベース化し、ソフトウェアGene Viewを開発した。
