研究概要

T細胞上のCD28/CTLA4とそのリガンドである抗原提示細胞上のCD80/86(CD28経路)はT細胞の活性化において重要なcostimulatory signalを与え、CD28/CTLA4とそのリガンドとの結合を阻害することにより抗原特異的な免疫学的不応答を誘導できることが知られている。最近の報告ではさらにCD28経路はT細胞依存性の抗体産生においても重要な働きを担うことが示されており、その阻害は抗原特異的抗体産生を抑制することが報告されている。われわれはCD28/CD80CD86の経路を介した免疫応答の制御機構に関し、以下の実験系でその分子の果たす役割を明らかにした。1)マウスの異所性心移植の系で抗CD80と抗CD86の併用投与で抗原特異的寛容の導入に成功し、以前に報告した抗LFA‐1/ICAM‐1抗体で誘導維持される寛容とはその細胞性機序が異なっていることを明らかにした。2)慢性の免疫異常モデルのNZB/NZW F1を用いCD80/CD86からのシグナルを阻害したための効果を調べ、慢性に進行する自己免疫病変が阻止出来得る結果を得た。
