未分化造血幹細胞の特性の総合的解析と臨床応用へのアプローチ

研究概要	これまでの研究を続行し,下記の成果をあげるとともに最終年度としての取りまとめを行った. 1.臍帯血に含まれる未熟な造血幹細胞を集め,造血因子とともに支持細胞上で長期培養を行い,未熟な幹細胞CD34^+細胞をc‐kit発現の有無により分画し妊娠週例による変化を観察した. 2.正常ヒト造血幹細胞の分化に伴うFas抗原発現とアポト‐シスとの関連を検討した.CD34^+Fas^-細胞は最も未熟な幹細胞に含まれ,その後成熟に伴ってCD34^+Fas^+細胞が増加した.この結果アポト‐シスを通じた造血の調節機構がうかがわれた. 3.我々の樹立した未分化な造血因子依存性巨核球細胞株(UT‐7)をエリトロポエチン(Ep)と長期培養して生じたEp依存性株(UT‐7/EPO)についてEp添加後のシグナル伝達等を検討し,新たにPLCγ1の系が活性化されていることを見出した.またリイトカイン無添加培養により因子非依存性亜株をつくり,チロシンホスファターゼ発現による増殖機構を検討した.更にトロンボポエチン依存生の亜株を樹立しこれ迄の亜株と比較し,巨核球産生機構を検討した. 4.Ep受容体には全長型(F)と短縮型(T)があり,赤芽球の成熟に伴いTからFへと消長しアポト‐シスを制御し造血の調節を行うと考えられる.この際c‐jun癌原遺伝子が関与することを示した. 5.臨床への応用の検討.(1)G‐CSFを投与後末梢血から採取したCD34陽性細胞の特性を調べ,幹細胞移植への応用を検討した.(2)赤白血病では赤血球に特異的な転写因子GATA1の発現が他の骨髄性白血病とは異ることを見出した.
