研究概要

マイクロニードルによる単一アクチンフィラメントのナノメーター操作技術を用いて、ミオシン1分子の発生する変位、力を直接計測する事に成功した。アクトミオシン分子モーターがATPの化学エネルギーを力学エネルギーに変換するメカニズムを解明するためには、1分子レベルでATPの分解反応検出する技術を開発しなければならない。
そこで、1分子の蛍光色素を水溶液中でリアルタイムでイメージングする蛍光顕微鏡法を開発した。この顕微鏡を使って、1分子のミオシンによる蛍光性のATPアナログ1分子の加水分解反応を直接イメージングすることに成功した。
