超効率的酸性触媒による合成反応の研究

研究概要	1.五塩化アンチモンと過塩素酸リチウムから成る触媒の存在下、芳香族化合物に酸無水物を作用させると、フリーデルクラフツアシル化反応が円滑に進行し、対応する芳香族ケトンが高収率で得られることを見いだした。また、触媒として四塩化ケイ素と過塩素酸銀を合せ用いると、カルボン酸または対応するトリメチルシリルエステルを直接アシル化剤として使用できる。反応はP-トリフルオロメチル安息香酸存在下で混合酸無水物を経由して進行し、穏かな条件下高収率で芳香族ケトンが得られることを明らかにした。 2.触媒量の四塩化チタンとトリフルオロメタンスルホン酸銀またはスズ(II)トリフラートとP-トリフルオロメチル安息香酸無水物の存在下、ほぼ等モル量のカルボン酸トリメチルシリルエステルとアルコールのトリメチルシリルエーテルから、対応するカルボン酸エステルが高収率で得られることを明らかにした。この反応は非常に穏やかな条件下進行し、クロトン酸、セノシオ酸などのエステル化においても、二重結合の異性化や移動を伴うことなくエステルを得ることができる。また、チオエステルやフェノールエステルの合成にも適用できることを見いだした。 3.リボースの1,2-シスおよび1,2-トランスグリコシル化反応、グルコースの1,2-トランスグリコシル化反応、これまで非常に困難とされ合成例のなかったアラビノースの1,2-シスグリコシル化反応、2-アミノ-2-デオキシグルコースおよびガラクトースの1,2-トランスグリコシル化反応など、数々の立体選択的グリコシル化反応を開発した。これからの反応はいずれも穏かな条件下進行し、触媒量の活性化剤を用いることで高い選択性を実現している。一方、これらの反応における触媒サイクルも明らかにし、溶媒、反応温度、対イオンの効果など、新しい知見を得ることができた。
