研究概要

本研究計画では、細菌の環境への応答機構を包括的に理解するために、分子遺伝学的・分子生物学的解析基盤が確立されている大腸菌を対象として、浸透圧や高塩ストレスに応答した細胞内情報伝達機構と遺伝子発現機構の解析に焦点を絞り、その分子機構を明らかにすることを目的として研究計画立案及びその遂行を目指した。特に本年度は、浸透圧に応答したK^+-イオン透過系に関与するK^+-ATPaseの発現制御に焦点を絞り解析し、以下の点を明らかにした。
(1)、K^+-ATPaseをコードするKdpABC遺伝子群のプロモーター構造を明らかにし、このプロモーターの発現が浸透圧ストレスに応答して転写レベルで制御されていることを示した。
(2)、Kdpプロモーター発現の調節因子であるKdpD及びKdpEタンパク質を同定・精製し、その生化学的性質を解析した。
(3)、KdpDは膜局在性の侵透圧センサーと想定され、自己リン酸化能及びKdpEに対するリン酸化能を有することを証明した。
(4)、KdpEはDNA結合性の転写活性化タンパク質であり、Kdpプロモーター上流に結合することを証明した。
(5)、KdpEはKdpDによりリン酸化され、その結果、Kdpプロモーター領域への結合能、及び転写活性化能が著しく上昇することが明らかとなった。
(6)、Kdpプロモーター上流領域のKdpE結合部位には特徴的な湾曲構造が存在することが明らかとなった。
以上の結果より、大腸菌における浸透圧ショックに応答したKdpABC遺伝子群の発現制御の全貌を分子レベルでほぼ明らかにすることができた。今後は、KdpEによる浸透圧検知、情報変換の分子機構に焦点を当てて解析を進め、本一般研究(B)の当初の目的の達成を目指す予定である。
