研究概要

本研究は、肺胞II型細胞におけるリン脂質代謝動態の調節におけるサーファクタント・アポ蛋白、特に親水性糖蛋白質のSP-AとSP-Dが関与する分子機構を解明することを目的とする。平成4年度の研究計画は概ね達成された。新たに得られた知見は下記の通りである。
1.SP-A及びSP-Dの特定脂質結合性とリン脂質代謝動態への関与
TLCリガンド法を用いて、SP-Aはジパルミトイルホスファチジルコリン(DPPC)、SP-Dはホスファチジルイノシトール(PI)に特異的に結合することを見出した。サーファクタント主成分のDPPCは、SP-Aと特異的に結合することにより、肺細II型細胞へのDPPCの取り込みを促進し、また細胞内でのラメラ封入体への移動を促進することを示した。PIはSP-Dへの結合リガンドであるが、同じリポソーム中のDPPCがSP-Aと結合した時、SP-D・リン脂質複合体がSP-Aの生物活性を抑制する可能性が示唆された。
2.SP-Aの分子構造と受容体への結合及び生物活性発現の解析
SP-AのC末端領域が、SP-Aの生物活性の発現に関与することを示した。SP-A受容体への結合及び生物活性発現を阻止し得る抗SP-A単クローン抗体(PE10)のエピトープ、つまりSP-Aの機能発現に関与する部位は、C末端領域のGly-202からMet-208に存在することを明らかにした。 SP-Aの受容体結合はCa依存性であるが、SP-AのCa結合部位はC末端領域でありSP-A1モルにCa2モルが結合することを示した。
3.SP-A受容体の性状の解析
ヒトおよびラットSP-A単クローン抗体に対する抗イディオ抗体を作製した。この抗イディオ抗体の肺胞II型細胞形質膜への結合は、SP-Aによって抑制されたので、抗イディオ抗体の認識する分子は、SP-A受容体であることが示された。
