研究概要

本研究は、肺胞H型細胞におけるリン脂質代謝動態の調節におけるサーファクタント・アポ蛋白、特に親水性糖蛋白質のSP-AとSP-Dが関与する分子機構を解明することを目的とし、遂行された。平成4、5年度に得られた成果を要約する。
1.SP-A,SP-Dの特定脂質結合性とリン脂質代謝動態の調節
SP-Aはジパルミトイルホスファチジルコリン(DPPC)と特異的に結合し、肺胞H型細胞によるDPPCの再利用効率を著るしく増大することを見いだした。一方、SP-Dはホスファチジルイノシトール(PI)と特異的に結合し、SP-Aによるリン脂質代謝動態の調節と拮抗することを明らかにし、両アポ蛋白質のリン脂質代謝動態調節の関与を示した。
2.SP-Aの生物活性発現に関与する分子構造とCa結合性
SP-AのC末端領域、特にGlu^<202>からMet^<207>までの6残基が、SP-Aの生物活性(肺胞H型細胞受容体への結合を介するサーファクタント・リン脂質の分泌抑制)の発現に重要であることを証明した。この中にCysがあり、ジスルフイド形成がSP-Aの生物活性発現に必須であることから、C末端領域が二つのジスルフイド結合で球状構造となり、その外表面にGlu^<202>-Met^<207>が位置し、受容体と結合すると結論した。一方、SP-AのCa結合部位はC末端で2モル結合し、このCa結合はSP-Aの生物活性発現に必須であった。
3.SP-Aに対する受容体分子
SP-A単クローン抗体に対する抗イデオ抗体の肺胞H型細胞への結合は、SP-Aによって阻害されたので、抗イデオ抗体によって認識される分子を検索した。その結果、肺胞H型細胞形質膜に存在する30kDa蛋白質が同定された。
