天然物由来の新規医薬品リード化合物の探索研究

研究概要	我々は天然資源から生物活性をもつ化合物の探索研究を長年にわたって続けてきたが、平成4年度から天然資源としてその種類も多く、全般的に成分探索が不十分であると思われる担子菌、子襄菌を中心に、1.抗腫瘍活性(細胞毒性物質)、2.酵素活性阻害物質(ここでは5-リポキシゲネース)、3.グルタミン酸アゴニスト、アンタゴニスト活性を指標としたスクリーニングを行い、新規活性化合物を単離し、その化学構造を決定し、活性と化学構造との関係を追求する研究に着手したが、本年度の研究実績の概要を以下に示す。(1)抗腫瘍活性:P388マウス白血病細胞に対する細胞毒性を指標にしたスクリーニングにより、クロアザアワタケ(Xerocomusnigro maculatus Hongo)の水溶性画分より得た化合物の構造研究を行ったところ、1-β-D-アラビノフラノシルシトシンであることを確認した。ニセクロハツ(Russula subnigricans Hongo)より得られた新規細胞毒性化合物(ルスフェリンと命名)の構造決定を行った。本化合物は塩素原子を4個含んだトリフェニルエーテル構造を有することを機器データより推定、合成により確認した。化学構造の修飾により活性は劇的に増強することを認めた。カノシタ(Hydnum repandum Fr.)より得た化合物の構造は分子内にアセチレン基とエポキシド基をそれぞれ2個有する化合物であると推定し、合成によってその立体配置をも含めて確認した。(2)5-リポキシゲナーゼ阻害物質として、クロカワ(Boletopsis leucomelas)より単離した化合物の構造を決定した。この化合物は既知の阻害剤AA-861とほぼ同等の阻害活性を示し、そのIC_<50>値は0.35μMであった。コウモリタケやニンギョータケからも阻害作用を示す化合物を単離した。(3)グルタミン酸アゴニスト、アンタゴニスト活性を示す化合物をドクササコ(Clitocybe acromelalga)より数種単離し新規カイノイドであることを確認した。
