ラン藻と葉緑体の転写後制御因子に関する研究

研究概要	葉緑体遺伝子の転写後制御に関与すると考えられるRNA結合蛋白質(RNP)の起源を明らかにするため、単細胞性ラン藻Synechococcus PCC6301から核酸結合蛋白質を分乱しその遺伝子構造を解析した。本研究で以下のことを明らかにした。 1)相同な2種類の核酸結合蛋白質(12RNP1と12RNP2と命名)の存在を蛋白質分析と遺伝子の構造解析の結果明らかにした。 2)12RNP1は110アミノ酸残基、12RNP2は99残基からなり、どちらも82残基のRNP-CSタイプのRNA結合ドメイン(CS-RBD)をもち両者の相同性は64%であった。葉緑体RNPのそれとは40〜52%と高い相同性をもつが、動物と酵母のRNA結合蛋白質とは18〜34%の相同性であった。 3)12RNP蛋白質は原核細胞で初めて見出された典型的なCS-RBD蛋白質である。 4)12RNP1遺伝子と12RNP2遺伝子は、530ntのモノシストロニックmRNAとして転写され、mRNAの蓄積量は両者で20倍以上異なる。 5)12RNP蛋白質は単鎖DNA、ポリ(G)、ポリ(U)鎖に特異的に結合するが、二本鎖DNA、ポリ(A)、ポリ(C)鎖には結合しないことをin vitro核酸結合実験で明らかにした。 6)ラン藻12RNPと葉緑体RNPおよび植物細胞のグリシンリッチRNA結合蛋白質(RGP)との進化的関係について考察した。
