植物細胞分化の不可逆過程の解析

研究概要	ヒャクニチソウ葉肉細胞から管状要素(道管・仮道管細胞)への細胞分化過程には変換点が存在(分化誘導後48時間目頃)し、この時期を境に、分化が可逆的な過程から不可逆過程に移行する。今回の研究ではこの不可逆過程を生化学・分子生物学的に解析した。まず、二次細胞壁形成の分子マーカーの単離を試み、二次細胞壁のパターン形成に関与するチューブリン遺伝子、二次細胞壁特異的なリグニンの合成に関与する遺伝子であるphenylalanine ammonia-lyase(PAL)と細胞壁結合peroxidaseの遺伝子(cDNA)を単離した。このうちperoxidase遺伝子ZPO-Cは、管状要素分化特異的に、しかも、リグニン合成の直前に一過的に発現することが示され、不可逆過程でのみ発現することが明らかになった。また自己分解のよいマーカーとなるシステインタンパク質分解酵素cDNAの単離に成功した。この遺伝子を単離し、細胞壁形成に関連する遺伝子との発現制御の比較をすることで、不可逆過程を引き起こす仕組みに迫れるものと期待される。次に、分化の変換点で発現する遺伝子の単離を試みた。そして、そのような遺伝子のcDNAクローンとして、遺伝子TED2-4を48時間目の培養細胞由来のcDNAライブラリーから単離した。in situ hybridizationにより、これらの遺伝子の植物体での発現を調べたところ、TED3が道管に分化する能力を持った細胞に、TED4が木部に分化する能力を持った細胞に、またTED2が木部、師部両方に分化する能力を持った細胞で発現することが分かった。そして、葉肉細胞から管状要素への分化系において、細胞分化能力は木部・師部→木部→道管(管状要素)へと次第に制限されていくと推定された。
