研究概要

1.ニワトリの網膜には、赤、緑、青、紫に特に感受性の高い4種の錐体視物質 Chicken Red,Green,Blue,Purpleがある。Chicken Blueを単離し、低温分光法を行った。他の視物質と同様、メタIとII中間体が存在した。またそれらの崩壊の時定数は、Chicken RedやGreenとほぼ同じで、ロドプシンに比べて早く崩壊することがわかった。2.赤色感受性錐体視物質であるアイオドプシン(Chicken Red)には、他の視物質と同様、メタI,II,III中間体の存在することがわかった。さらにメタI中間体は、メタII中間体へ崩壊すると共に、Chicken Redに暗再生することがわかった。3.ニワトリのロドプシンとChicken Greenとは、アミノ酸配列が互いに似ているので、特徴的に異なる3つのアミノ酸残基を選び、それらを置換したロドプシン変異体の遺伝子をサルの腎臓由来の株細胞(COS細胞)中で発現させることに成功した。現在、変異体及びその中間体の物理、化学的性質を調べている。4.ウシ網膜から精製したトランスデューシンのαサブユニット(Tα)のN末端グリシン残基には、4種類の脂肪酸がアミド結合している。これらの脂肪酸を識別する単クローン抗体を作成し、ウシの網膜切片と反応させたところ、1個の視細胞に4種のTαが存在していることを示唆する結果がえられた。5.ニワトリの松果体は概日リズムを司る時計と、これをリセットする光受容系の両者をもつ。松果体の光受容蛋白質をコードするcDNAを単離し、アミノ酸配列を決定したところ、他の視物質と約40%の相同性を示した。培養細胞系で発現させたところ、11-cis-retinalと結合し、光感受性を示した。(λmax:475nm)ので、“Pinopsin"と命名した。
