中胚葉分化誘導因子による細胞分化の決定と形態形成に関する分子生物学的研究

研究概要	平成5年度の研究実績は以下の通りである。まず、アクチビン(10ng/ml)とレチノイン酸(RA)をいろいろな濃度で混合すると、細胞分化の質の変化がおこる。RAを10^<-6>Mとの混合では著しく筋肉の分化がおこり、RAを10^<-5>Mでは非常に高率に腎管を分化誘導がおこった。未分化細胞に既知の物質(アクチビンとRA)を与えて腎管を分化誘導したのはこれが初めてである。次に、ツメガエルの初期胚のCa^-、Mg^-free溶液中に入ると、各割球にバラバラになる。その時、背側割球と腹側の割球と分離して取り出して、各々、別々にアクチビン処理する。すると、背側割球には筋肉や脊索を分化し、腹側割球は血球や体腔内上皮を分化する。つまり、8細胞期の割球にはすでに背側と腹側との割球にプレパターンが存在する。また、筋肉分化を分化誘導するアクチビンのタンパク質の精製を行った。まずアクチビンとファリスタチンを精製・単離する方法を確立した。N末端のアミノ酸配列から、3種類のアクチビンA、AB、B、およびフォリスタチンをタンパク質としてはじめて精製することができた。フォリスタチンについてはN^-末端尾アミノ酸配列の決定も行われた。ツメガエルのフォリスタチンと他の哺乳類動物との比較検討を行った。更に、ツメガエルの初期胚そのものから胚発生で筋肉分化をひきおこすと考えられるアクチビンの単離を試みた。ツメガエルの非常に早い初期胚(stl-5)1万個から、上記の方法をつかってアクチビンとフォリスタチンが存在することがわかった。アクチビンBが他のアクチビンAまたはABよりも少し多く、フォリスタチンはアクチビンの約40倍の量が存在し、1個の卵当り、アクチビンは約1pg、フォリスタチンは約40pgがタンパク質の形で存在している。
