遺伝子発現制御因子間の相互作用による機能調節

研究概要

細胞増殖の制御に関与するいくつかの転写制御因子を取り上げ、蛋白質間相互作用による機能調節について解析した。
1)mybがん遺伝子産物(Myb)の中央部にロイシンジッパー構造が存在し、この部位にインヒビターが結合し、Mybの機能を抑制することを明らかにした。インヒビターをcDNAクローニングにより同定し、これをLassin(leucine zipper-associated inhibitor)と名付けた。LassinがMybに結合すると、MybはDNAに結合できなくなることが明らかとなった。またMybはロイシンジッパーを介してホモダイマーを形成し。ホモダイマーはDNAに結合できないので、Mybが過剰量存在する時は、Myb自身がインヒビターとして機能するnegative autoregulationの機構も存在することが示された。
2)skiがん遺伝子産物(Ski)は核内に局在することから転写因子として機能することが推定されているが詳細は不明である。本年度はSkiがホモ三量体やSki関連遺伝子産物Snoとヘテロ三量体を形成する能力を持つことを明らかにした。
