ビブリオ属菌の産生するいわゆる溶血毒の病原的意義の解明

研究概要	ビブリオ属菌の中でも代表的な病原菌である腸炎ビブリオの耐熱性溶血毒(TDH)およびO1コレラ菌の産生するエルトール型溶血毒(Hly)を中心に研究をすすめた。165個のアミノ酸から成るTDHの種々な位置でアミノ酸置換をおこさせ、変異毒素を作成した。その結果、これまで毒素の化学修飾法等でわれわれが見出したTrp^<65>とGly^<90>の他にGly^<62>、Thr^<67>、Gly^<86>、Glu^<116>、Glu^<138>のアミノ酸残基がTDHの活性発現に重要なこと、特にGLy^<62>とTrp^<65>の変異は活性に大きな影響を示すにもかかわらず、標的細胞への結合能は保持していた。一方^<125>Iで標識したTDHを用いて結合実験を行ったところ、TDHに抵抗性(溶血しない)ウマ血球も他の血球と同様に結合することが明らかになった。これらの結果は、TDHが作用を発揮するのに(1)結合、(2)膜破壊(孔形成)の少なくとも2つのプロセスが存在することを示唆する。さらにTDH(あるいは類似毒素TRH)の遺伝子を欠失させた変異株を作成したので、病原性の変化の有無を検討中である。コレラ菌Hlyについては、この溶血毒が前前駆体(pre-protoxin)として生合成され、シグナルペプチド(pre領域)の切断の上に、菌体外で切断されるpro領域(アミノ酸132個から成る)を有するという特異な構造を有することを見出した。特にpro領域の存在は本毒素にユニークなもので、その役割をpro領域欠損変異株を作り調べたところ、Hlyの立体構造の保持の機能をになっていることが分かった。なお、Hly遺伝子欠失株の病原性は減弱していたので、Hlyの病原的意義も示唆できた。
