T細胞レセプター複合体の構造とシグナル伝達の解析

研究概要	1.CD3シグナルモチーフの役割:昨年度作製した、CD3ζモチーフを欠損させた変異ζ鎖(Zn)を有するTCR複合体の介する初期シグナル伝達系を解析した。抗原刺激によるIL-2産生にはZn細胞の変化がなかった事と相関して、IL-2産生に重要であるチロシンキナーゼZAP70やPLCγの活性化は同じように誘導された。この事はζシグナルモチーフが欠損していても他のCD3分子のモチーフでもIL-2産生系には、十分であることを示した。しかし、チロシンリン酸化蛋白については、90kd分子に代表される蛋白のリン酸化がZn細胞には欠損していた。Zn細胞がThy-1分子を介する刺激にも反応しない事実と共に、TCRシグナルにはζモチーフ依存性のものと、全てのCD3モチーフに共通なシグナルとが存在することが示された。 2.T細胞活性化におけるPI代謝系の役割:PLCγを直接活性化することのできるPDGFレセプターをT細胞に発現させて、そのシグナル伝達系の変化を解析した。発現されたPDGFRは、PDGF結合により、自己リン酸化を起こすと共に、多くの基質のリン酸化を誘導した。PLCγもチロシンリン酸化され、活性化され、結果として細胞内Ca^<2+>上昇が起こったが、IL-2産生は誘導しなかった。しかし、抗原刺激に伴うIL-2産生を著名に増強した。これらより、PI代謝系を活性化しただけではIL-2産生に至らず、TCRからのチロシンリン酸化シグナルとの相互作用が必要であることが判明した。 3.ζ鎖とダイマーを形成する新たな分子:正常T細胞にも存在して、分子量薬12kdでζ鎖とヘテロダイマーを形成している分子を同定した。ペプチドマップでは、ζ鎖と類似しているが異なっており、T細胞ハイブリドーマ5×10^<10>から蛋白を精製した。最終的なアミノ酸配列は成功しなかったがN末端がブロックされていなかったことからも、ζ鎖ににた新たなファミリー分子であることが推測された。
