発生工学的手法を用いた免疫グロブリン遺伝子再構成と発現の制御機構の解明

研究概要	免疫グロブリン(Ig)H鎖遺伝子の再構成、転写制御を司るエンハンサー内に同定されたHE2/μB配列の役割を明かにするため、以下の研究を進めている。ジーンターゲティング法により、マウスES細胞内H鎖遺伝子のμB配列を除去するためのベクターを作製中であるが、μBと置換されたneo耐性遺伝子(neo)を除去する目的で用いるリコンビナーゼとその認識配列として、予定を変更してさらに効率のよいCre及びloxp配列を使用する。そして、2つのloxp間の配列の除去を薬剤選択できるように、HSVtk遺伝子をneoの隣に挿入する。上記ベクターをES細胞に導入し、G418耐性株の中から相同組換え体を選択する。後にキメラマウスでの解析を容易にする目的で、さらに高濃度G418で培養し、相同組換え変異のホモ接合体の作製を試みる。最後にCreの一過性発現によりneo及びtk遺伝子を両染色体上で欠失したES細胞を選択する。この変異ES細胞を、Ig遺伝子再構成機構の破壊されたRAG2欠損マウス由来の胚盤胞に導入し作製したキメラマウスでは、発生するB細胞はすべてES細胞由来となるので、Ig遺伝子再構成や転写を解析することが比較的容易であると思われる。一方、ジーントラップクローニング法に関しては、ジーントラップベクターのB細胞への導入効率が低いこと、HSVtk遺伝子の発現によるGANC感受性化が完全でないこと等の理由で、当初予定したシステムがうまく作動していない。しかし、この方法はX染色体上の転写活性化因子を単離するのに特に有効と考えられ、XidのようにIg発現の異常がX染色体に連鎖する免疫不全マウスも存在することから、IgH鎖エンハンサーを活性化する機構の少なくとも一部がこの方法で解明できるものと期待される。今後、上記の問題点を解決し、研究を進めたい。
