モチリンによる空腹期収縮発現機構におけるセロトニンの末梢性、中枢性関与

研究概要	1.われわれが作製したモチリン及びセロトニン抗血清の特異性を検討した後、パラフィン包埋したイヌ十二指腸を用いてミラーセクションを作製し、モチリンとセロトニンを免疫組織化学により染色した。その結果、モチリンとセロトニンの共存を示す細胞は全く見られず、2つの物質の共存は免疫組織化学的に否定され、モチリンとセロトニンの同一細胞よりの同時分泌はないと考えられた。 2.意識下犬の門脈中セロトニン、モチリン濃度を経時的に測定した結果、両物質の相関が見られなかった。これは、上記1の結果を支持するものである。 3.イヌにセロトニン合成阻害剤(PCPA)を経口投与すると数日後には空腹期収縮が完全に消失すると共に生理的濃度の外因性モチリンを投与しても空腹期収縮は惹起されなかった。また、血中セロトニン含有量はPCPA投与により徐々に減少し数日後には測定限界以下になった。この結果はモチリンの作用発現には中枢及び末梢(消化管)のセロトニンが必要なことを示唆していると考えられる。 4.上記の結果を踏まえて、モチリンの末梢性作用及びセロトニン、セロトニン3レセプターとの関係を調べるためにイヌ胃を生体より離断、人工血液にて潅流した。中枢から離断した胃は潅流液中へのモチリン投与により口側より肛門側に向かう蠕動運動が観察され、この運動はセロトニン3レセプターアンタゴニストにより完全に消失した。この結果より、モチリンに反応するセロトニン受容体は末梢(胃内)に存在することが証明された。しかし、この結果はモチリンの中枢性作用を否定するものではなく中枢の関与についてはさらなる研究が必要であると考えられる。
