気道粘膜下腺からの電解質分泌機序の解明

研究概要	1.ネコ摘出気管及びヒト手術肺気道から粘膜下腺を単離し、酵素処理で腺房細胞まで分散させ、これにパッチクランプ法を応用して164pS large conductance voltage-sensitive Ca^<2+>-sensitive K^+-channelsを基底外側膜に、培養ヒト粘膜下腺細胞の内腔側膜にCa^<2+>-sensitive Cl^--channels(4pS、15pS)を同定した。細胞内pH螢光指示薬BCECFを用いて、アンモニアパルス法によりネコ粘膜下腺にNa^+/K^+の存在を示した。 2.気道粘膜下腺の電解質分泌モデルとしては、IP_3受容体が内腔側膜直下に居在し、IP_3によるCa^<2+>遊離によりCl^--channelsを活性化させ、Cl^-分泌をもたらし、一方、K^+-channelsはryanodine感受性Ca^<2+>pool(CICR)からのCa^<2+>遊離にて制御されている。ウァバインやフロセマイドの膜電流に対する効果からして、Na^+/K^+-2Cl^-共輸送系及びNa^+/K^+ポンプはCl^-分泌に必須ではないが、その維持に必要である。 3.デキサメサゾン処理は電解質分泌を抑制させたが、その機序としてCl^--channels(CFTR)の膜への移動抑制が考えられた。 4.オゾン暴露によりもたられた過形成を示したネコ粘膜下腺での電解質分泌機能は正常と有意の差がみられなかった。
