研究概要	気管支喘息は、気道粘膜における慢性好酸球性炎症である。好酸球を特異的に活性化するサイトカインIL-5を産生する活性化T細胞は、アトピー型、非アトピー型(内因性)を問わず、気管支喘息の病態に共通して認められ、気道炎症の引き金の役割を担うことが、我々の研究から明らかにされた。ヒトT細胞のIL-5産生のプロセスを解明することは、新たな喘息治療の可能性をひらく観点から重要である。喘息患者の末梢リンパ球を抗原刺激にて樹立した。ダニアレルゲン特異的T細胞クローンは、抗原刺激、固相化CD3抗体、ホルボールエステル(PMA)+イオノマイシン(IOM)等の刺激により活性化され、増殖・サイトカイン産生を行った。IL-5産生は、抗IL-2中和抗体によって完全に阻害された。すなわちT細胞のIL-5産生はIL-2依存性であることがわかった。さらに、ヒトIL-2単独の刺激に反応して、IL-5産生がみられた、ヒトIL-5遺伝子上流約500bpのプロモーターノエンハンサー領域をルシフェラーゼ遺伝子に接続したレポータープラスミド、p IL-5 LUCに起因するルシフェラーゼ活性がIL-2刺激により誘導されたことから、IL-2レセプターを介するシグナルによりIL-5遺伝子転写が誘導されることも明らかとなった。IgE抗体が認められない喘息患者においても、ダニアレルゲン特異的T細胞クローンが樹立され、IL-5産生がなされることから、内因性喘息におけるダニアレルゲンの関与も推測される。
